Abstract
We developed a kind of virus-mimetic nanovesicle (Mel-VSVG-NA), which could react with tumor cells in a membrane fusion procedure (named as First Fusion), result by the function of Vesicular Stomatitis Virus Glycoprotein (VSVG) in acid microenvironment. Meanwhile, the melittin on Mel-VSVG-NA was transfer on the surface of targeted tumor cell, and thus execute hole-digging process on the tumor cell membrane, causing severe cell content loss and suppression of tumor growth. After the first-round treatment, a harmless ultrasound exposure was carried out, the further engineered vesicles were generated from the damaged cells. Afterwards, the produced vesicles were also confirmed to engineered with VSVG and melittin, which could further fuse with other tumor cells and induced another cell death (named as Second Fusion). These results indicated that Mel-VSVG-NA is a reliable advance of nano-treatment in ocular tumor therapy, and we bring in this concept of Hijacking- Integration-Vesicular amplification-Exocytosis mode as HIVE therapy.
